چکیده

We evaluated a one-tube multiplex real-time PCR targeting DNA of Schistosoma haematobium complex and S. mansoni in serum samples obtained at different German diagnostic centers. Simplex protocols for the detection multi-copy DNA-repeats Dra1 Sm1-7 were combined to new format. The was subjected full validation including evaluation real-life setting with travelers migrants. results compared those stool urine microscopy, serology, circulating cathodic antigen (CCA) rapid tests urine. Sensitivity specificity approaches analyzed using latent class analysis (LCA). LCA assessment indicated sensitivity 94.9% 98.4%, respectively, if serology included calculation, 100% 95.6%, not as parameter necessarily associated active infection. Agreement between procedures genus level fair (kappa 0.273) moderate 0.420) included. assay proved be highly reliable diagnosis schistosomiasis

Detection of Schistosoma mansoni and Schistosoma haematobium by Real-Time PCR with High Resolution Melting Analysis

The present study describes a real-time PCR approach with high resolution melting-curve (HRM) assay developed for the detection and differentiation of Schistosoma mansoni and S. haematobium in fecal and urine samples collected from rural Yemen. The samples were screened by microscopy and PCR for the Schistosoma species infection. A pair of degenerate primers were designed targeting partial regi...

Evaluation of real-time PCR assay to detect Schistosoma mansoni infections in a low endemic setting

BACKGROUND Schistosomiasis constitutes a major public health problem, and 200 million people are estimated to be infected with schistosomiasis worldwide. In Brazil, schistosomiasis has been reported in 19 states, showing areas of high and medium endemicity and a wide range of areas of low endemicity (ALE). Barra Mansa in Rio de Janeiro state has an estimated prevalence of 1%. ALE represent a ne...

A Schistosoma haematobium-Specific Real-Time PCR for Diagnosis of Urogenital Schistosomiasis in Serum Samples of International Travelers and Migrants

BACKGROUND Diagnosis of urogenital schistosomiasis by microscopy and serological tests may be elusive in travelers due to low egg load and the absence of seroconversion upon arrival. There is need for a more sensitive diagnostic test. Therefore, we developed a real-time PCR targeting the Schistosoma haematobium-specific Dra1 sequence. METHODOLOGY/PRINCIPAL FINDINGS The PCR was evaluated on ur...
